SUMMARY:Lebek: PUFFFIN - Illuminating Cellular Neighbourhoods in Model Syst
 ems of Development
CLASS:PUBLIC
DESCRIPTION:Speaker: Tamina Lebek\nInstitute of Speaker: Centre for Regener
ative Medicine\, Institute for Regeneration and Repair\, University of Edi
 nburgh\, UK\nTopics:\n\n Location:\n Name: MPI-CBG (MPI-CBG CBG Galleria
 II (VC))\n Street: Pfotenhauerstraße 108\n City: 01307 Dresden\n Phone

ells influence their neighbours is crucial for deciphering processes like
development\, homeostasis\, and disease. Recent advances have introduced f
luorescent protein-based methods to identify cellular neighbourhoods. I wi
ll present Positive Ultra-bright Fluorescent Fusion For Identifying Neighb
ours (PUFFFIN)\, a new neighbour-labelling system based on the secretion a
nd uptake of a super-positive fluorescent protein. PUFFFIN enables precise
and rapid identification\, isolation\, and profiling of neighbouring cell
s\, even capturing transient interactions. Its modular design allows colou
r-of-choice labelling and transgene integration\, making it highly versati
le. I will also demonstrate how we are using PUFFFIN to study coordination
of differentiation in mouse embryonic stem cells. This powerful tool offe
rs a simple\, sensitive\, and customisable approach for profiling cell-cel
l communication in diverse contexts\, paving the way for new insights into
  cellular interactions.
